Self-Run Policies and Information

We encourage researchers to use the Bio-Plex 200, LSR II, and FACScans independently. Self-run users enjoy many benefits, including 24-hour use of the facility, lower rates (FACScan only), and a better understanding of the capabilities and limitations of flow cytometry as a method.

Training

Bio-Plex 200 self-run users will take a two-hour training course, including machine and software information, sample preparation, and running your samples on the Bio-Plex.

LSR II Self-run users will take a two-hour training course which will include machine set up, software training, and your own sample analysis (It is recommended that LSR II users be self-run FACScan trained prior to LSR II training).

FACSCan Self-run users will take a two-hour intro training course. These classes cover the basics of flow cytometry theory, proper use of the FACScan, and facility policies.

Users will then learn to run their own samples in one-on-one sessions at the FACScan. Finally, they will take a practical exam to demonstrate their independence.

Users with prior experience using a FACScans, LSR II, or Bio-Plex may bypass the training course and go straight to one-on-one training, or to the exams.

Scheduling and Billing

Scan 2, Scan 3, the LSR II, and the Bio-Plex 200 may be scheduled for independent use during facility hours. All Machines may be used after hours.

As per facility policy, all time must be scheduled online. Users will be charged for time booked or time logged in, whichever is longer.

Purchase order users: We require a paper copy of each PO you use. If you are using a standing PO, you must keep track of your own usage.

Three-Strikes Policy

Any self-run procedures that are overlooked will count as one strike. Users who accrue three strikes will be limited to using the FACScans during facility hours. If you get three strikes beyond that, you will need to retake training before being able to run your own samples again.

Instrument Usage and Troubleshooting

Please use common courtesy when using the FACScans. Samples should be filtered as your last prep step. Clogging will cause delays for you and others.

Always check sheath and waste tank levels.

If someone is scheduled after you, log off the machine promptly.

If the FACScan or LSR II clogs, you may try BACKFLUSH(FACScan) or PRIME (LSR II), or try running 10% bleach or water through the sample intake. If these things do not work, let a staff member know. Do not take anything apart or blow air through the sample probe! Staff will do this if necessary. If your samples have caused the machine to clog once, please refilter them before continuing to run. You may need to book another appointment later in the day if someone is scheduled after you. The best way to avoid these problems is to filter your samples as the last step before running them.

Data Acquisition and Storage

We store all files in the DATE folders. Files should be named appropriately. You may save your own templates in your USER folder.

Do not save directly to storage media. Do not work directly from storage media. Files which are in use must be saved to the hard drive.
